The embryogenic callus was transformed under optimum transformation conditions. After the second round of Hyg screening, resistant yellow callus in 31.58% of the total transformed callus was selected and half was used to induce somatic embryos. Finally, histochemical staining of callus and somatic embryos for GUS gene expression determination and a hand-held lamp (LUYOR 3415RG, Shanghai, China) were used to investigate the expression and resulting fluorescence signals of GFP in resistant transgenic callus and somatic embryos.
